ABSTRACT

Objective To study the effect of an immuno-targeted therapy drug Herceptin on apoptosis and cell cycle of overexpressing HER2 breast cancer cells. Methods Breast cancer cell line SKBR-3 was treated with Herceptin at most effective dosage and exposure time were selected by MTT assay. Then the apoptotic features, the apoptotic rate and the change of cell cycle of breast cancer cells were detected by fluorescent microscope(FM),laser confocal microscope(LCM),scanning electron microscope(SEM),transmission electron microscope(TEM)and flow cytometry(FCM).Results With the treatment of Herceptin, the visible apoptosis features of SKBR-3 cells were observed by FM,LCM,SEM and TEM.Compared to the control cell group, the rate of initial apoptotic cells detected by FCM with Annexin V/PI staining increased significantly(P

ABSTRACT

Objective To investgate the changes of bone mineral density (BMD) and biochemical markers of bone metabolism in elderly male patients with osteoporosis (OP) . Methods BMD and bone mineral content (BMC) of the lumbar spines (L 2~4 ) and the biochemical markers of bone metabolism in serum and urine were measured. Results BMD and BMC in OP group was reduced by 21.6% and 25% respectively vs that in control group. Bone formation indicators of serum levels of ALP and BGP in OP group were increased by 25.4% and 222% respectively vs that in control. At the same time, bone resorption indicators of urine Hop/Cr and NTX/Cr ratios were significantly increased by 22.6% and 223% in OP group than that in control. Serum T level of OP group was lower than that in control group and serum 25 OH D3 was below the normal value in both groups. Conclusions BMD and BMC of the lumbar spines (L 2 4 ) are the main basis for diagnosing the osteoporosis in men. Some elder male patients with osteoporosis are due to high turnover of bone metabolism. Androgen plays an important part in maintaining the bone mass. Vitamin D deficiency in elder men is the main reason occurring osteoporosis.

ABSTRACT

Objective To observe the toxic effects of mercuric chloride (HgCl2) on the productive function of male mice with lower dosage exposune. Methods 4 week-aged male ICR mice were randomly divided into 3 exposure groups, and control group. The 3 exposure groups were treated with doses of 0.25, 0.50, 1.00 mg/kg HgCl2 by peritoneal injection respectively, one time per 3 days, 10 times in total. After exposure to HgCl2 for 50 days, the male mice were mated with female mice non-exposed to HgCl2 in a ratio 1∶2. The pregnant rate, number of pups whelped per group, body weight of offspring, testis index, sperm count, sperm motility rate, abnormal sperm rate were observed. Results The pregnant rates were 100%, 100%, 83.33% and 66.67% for control group, 0.25 mg/kg group, 0.50 mg/kg group and 1.00 mg/kg group respectively during 1-week conception, 100%, 100%, 83.33% and 75% for above corresponding groups respectively during 3-weeks conception respectively. The pregnant rate of 1.00 mg/kg group was significantly lower than that of control during 1-week conception (P
